MitoEAGLE Short-Term Scientific Mission

Work Plan summary
I was introduced to the field of mitochondrial physiology during my pre-graduate medical studies in the academic year 2016/2017 at the Department of Physiology, Faculty of Medicine in Hradec Kralove, Charles university Czech Republic. :::: I co-operated with Dr. Ondrej Sobotka on a project focused on respiration of liver mitochondria during the development of non-alcoholic fatty liver disease (NAFLD). We performed High Resolution Respirometry (HRR) using Oroboros O2k Oxygraph. I found this project very interesting, especially due to the steep increase in the incidence of NAFLD throughout the western world. Also, it has been shown recently, that mitochondrial functions are affected in a number of other pathologies including obesity, diabetes, Alzheimerโ€™s disease and cancer.
As a practicing doctor in nuclear medicine, we work with radiopharmaceuticals as diagnostic tools for various organ systems including musculoskeletal, cardiac, pulmonary and endocrine. It is a powerful and prospective method for early diagnoses of cancer metastases, evaluation of cardiac ischemia and detection of pulmonary embolisms.
Mitochondrial-targeted-molecular imaging is a technique that uses radiopharmaceuticals for functional imaging of targeted tissues. This method is deployed in the Myocardial Perfusion Scintigraphy which is widely used in the clinical setting for examining cardiac function, detecting coronary artery disease and differentiating between ischemic and necrotic myocardiocytes. Research on mitochondrial-targeted imaging include studies which use PET imaging in monitoring the Mitochondrial Complex I in Parkinsons disease models and recently, the use of PET in assessing mitochondrial membrane potential in lung cancer. I view this connection between nuclear medicine and mitochondrial pathophysiology as a potential discovery-translation-application pipeline in both fields and opportunity for further and deeper exploration of mitochondrial functions.
I believe that for me STSM will be a very beneficial and pivotal opportunity to broaden my theoretical knowledge of mitochondria, further train my practical skills using the available technologies at Oroboros Mitochondrial Physiology Lab as well as learning new techniques and training on the latest DatLab software. This experience will surely provide me with insight and improved proficiency to make sound scientific judgement while approaching research in my field and it will allow me further communication within this MitoEAGLE scientific community and more future participation.
