Are you planning to perform several measurements in one day using the same O2k-chamber?

To avoid any possible contamination by inhibitors in the subsequent measurements, the O2k-chamber needs to be cleaned each time with the standard protocol AfterUse or AfterUse_inhibitors.

Does your polarographic oxygen sensor (POS) need a deep clean?

A POS with a very dark anode might need several rounds of ammonia cleaning, possibly including prolonged exposure overnight to restore performance specifications.

Add catalase when measuring Complex IV (CIV) activity in permeabilized fibers (pfi)

Pfi have a high content of iron-containing proteins, which accelerate the autoxidation of tetramethyl-p-phenylenediamine (TMPD) in the CIV assay. Catalase prevents the accumulation of H₂O₂ generated by this autoxidation.

Do you observe simultaneous fluctuations in the O₂ slope neg. and the Peltier power?
