Description
Mammalian superoxide dismutase (SOD) exists in three forms, of which the Mn-SOD occurs in mitochondria (mtSOD, SOD2; 93 kD homotetramer) and many bacteria, in contrast to the Cu-Zn forms of SOD (cytosolic SOD1, extracellular SOD3 anchored to the extracellular matrix and cell surface). Superoxide anion (O2•-) is a major reactive oxygen species (ROS) which is dismutated by SOD to oxygen and H2O2.

Application in HRR
- Add SOD for conversion of O2•- and measurement of H2O2 with Amplex ultrared.
- SOD: Superoxide dismutase from bovine liver, Sigma S8160, 15 KU, store at -20°C; MW = 32.000,00 g/mol
- Preparation of 5000 U/mL stock solution (dissolved in H2O)
- Weigh 2 mg of superoxide dismutase.
- Dissolve in 1134 µL H2O.
- Divide into 30 µL portions into 0.2 mL Eppendorf tubes.
- Store frozen at -20 °C in the desiccator.
