MitoEAGLE Short-Term Scientific Mission

Work Plan summary
My name is Manuela Passrugger and I studied Biomedical Science at the University of applied Science in Salzburg (FH Salzburg). I am writing this letter to apply for the Short-Term Scientific Mission (STSM) covered by MitoEAGLE COST Action. Since December 2016 I am working for Oroboros Instruments in Innsbruck/Austria. I am working with standardized Substrate-Uncoupler-Inhibitor Titration (SUIT) protocols and am trained very well in essential quality control (QC) procedures in our Laboratory. In science, frequently, we cannot reproduce many results which are already published, that represents a problem for the scientific progress. QC is a key factor to obtain reproducible data. QC starts way before the experiment with the maintenance of the instrument e.g. service of parts, calibrations and experimental background measurements. The first step to get comparable data is a good running instrument. Afterwards, an adequate design of the protocols (sample preparation, SUIT protocols) are needed to obtain optimal results. In the context of MitoEAGLE, I could contribute in the optimisation of SUIT protocols and also get to know different cell models (HUH7/ C2C12/ human lymphocytes) as they currently use in the School of medicine laboratory in Belgrade, Serbia. Furthermore, we would have the possibility to discuss and share QC criteria (implement standards in respirometric OXPHOS analysis, applied in comparative mitochondrial physiology, mitochondrial medicine, and clinical diagnosis). It would be a great start of making high resolution respirometry (HRR) measurements by using Oxygraph (O2k) in two different labs comparable, as a first step approaching comparability in many labs and finally in literature. This STSM would be an important possibility on one hand to connect Austria to an inclusiveness target country (ITC) as Serbia, opening future collaborations and on the other hand for me to broaden my horizon and gain more skills. I would be very grateful if you consider my application and give me the chance to participate in the previously described Short-Term Scientific Mission MitoEAGLE.
