Abstract: Influenza viruses have been responsible for highly contagious acute respiratory illnesses with high mortality, mainly in the elderly and immune compromised patients, which encourages the development of new drugs for the treatment of human flu. The biotherapics are medicines prepared from biological products, which are compounded according to homeopathic procedures indicated for infectious diseases with known etiology. The purpose of the present study is to develop biotherapics prepared from the intact and inactivated influenza A virus (A/Aichi/2/68 and A/Victoria/3/75), at 12DH and 30DH potencies, and evaluate its in vitro, pre-clinical and clinical effectiveness. The intact biotherapic was prepared according to Roberto Costa’s technique and the ultrastructural aspects of viral particles prepared at the potency of 1DH in two solvents (water and alcohol 70%) were evaluated. Transmission electron microscopy images indicated that the viral particles remained intact and completely lysed in the presence of water and alcohol 70%, respectively. The in vitro experiments showed alterations in important enzyme activities and in the maximum respiratory capacity when MDCK were treated with intact biotherapic 30DH. In contrast, biochemical and cell alterations were not detected in the macrophages tested (J774G8 e RAW 264-7). The preclinical study was carried out in Balb-c mice, and the results indicated that the intact and inactivated biotherapic at 30DH was not able to induce pathogenetic effects and/or immune alterations, when the animals were challenged with influenza antigen, after 21 days, using the biotherapic ad libitum. In contrast, in the second set of experiments with intact biotherapics 12DH and 30DH, some important cell alterations induced by 30DH were detected, namely: B and non-B cells increased, B2 cells decreased, B1 and CD4+ increased. These alterations indicated that the treatment with biotherapic 30DH was able to stimulate the immune response when the animals were challenged with influenza antigen. The clinical trial done in partnership with Roberto Costa Institute and Brazilian Public Health System in Petropolis/RJ proved that homeopathic medicines (intact biotherapic 30DH and IRA) have a prophylactic effect against flu and acute respiratory infection symptoms , when compared to placebo. This clinical trial, pioneer in the literature, evaluated a significant number of children (600) ranging from 1 to 5 years of age, by a triple blind, randomized and placebo controlled protocol, bringing robustness and reliability to this investigation. The set of results obtained in this study provide strong evidence of the therapeutic and prophylactic potential of the intact biotherapic 30DH against infections caused by influenza viruses.
