Abstract: Besides of the 23 chromosome pairs located in the nucleous (nDNA), the human genome also includes many DNA molecules placed in mitochondria and known as mitochondrial DNA (mtDNA). mtDNA encodes 13 subunits very important for the oxidative phosphorylation system (OXPHOS) and RNAs required for its expression.

Because of its location and genetic particularities, mtDNA accumulates mutations faster than nDNA. Many of these mutations result into a serious disease. The construction of cellular models are the key to study these mutations.

In his work we used the cell line HepG2, beacause its utility in in vitro studies. We studied their differentiation into mature hepatocytes and tried to remove its mtDNA to build transmitochondrial cell lines. Keywords: HepG2 cells
